How to seal the PCR plates?

Plastic PCR plates are available in two ways of sealing, optical caps and optical caps.

Optical cover:

1. Place the plate on the support base.

2. Remove the white backing and place the lid on top of the board, adhesive side down, inside the raised edge. The cap comes with a square plastic applicator.

3. Secure the board by attaching the board and support base to the edge. Rub the flat side of the applicator back and forth along the long side of the plate.

4. Rub the flat side of the applicator back and forth along the short side (width) of the plate. 

5. Use the edge of the applicator and seal around the edge of the plate and both horizontally and vertically between the wells. Make sure there is a good seal around all wells and the outside edges of the board.

6. Using the applicator tool, press down on each end of the cap and remove the white strip from the perforation. Be careful not to pull up on the lid.

7. Check the top of the board to verify that the cover is in good contact with the board, especially along the edges.

After sealing the board, lift it up and inspect the bottom. Briefly centrifuge the plate. Check the plate for empty wells, droplets adhering to the walls of the wells, and air bubbles at the bottom of the wells. Centrifuge again if needed. Mark any empty wells on the plate loading guide.

Plates can be marked along the edge of the plate, but do not write on the surface of the plate as this will interfere with fluorescence excitation and detection. Also, don't write on the bottom of the plate. The ink can be transferred to the module in the real-time PCR instrument. Significant block contamination of colored or fluorescent substances can adversely affect real-time PCR data. Some plates have barcodes that can be configured and scanned to help identify the plates.

Cap strips:

1. Place the board on the support base 

2. Align the cap strips with the corresponding wells on the 96-well plate

3. Use the cap installation tool to seal the cap strip

It is best not to seal the cap by hand. Even with gloves on, they can cause interrupted fluorescence readings if hands or gloves become contaminated.
