What is a nucleic acid extractor?

Nucleic acid extraction system is an instrument that automatically completes the nucleic acid extraction of samples by applying the supporting nucleic acid extraction reagents. Widely used in disease control centers, clinical disease diagnosis, blood transfusion safety, forensic identification, environmental microbial testing, food safety testing, animal husbandry and molecular biology research and other fields.

What is the magnetic bead method?

Using magnetic beads as the carrier, using the principle that magnetic beads adsorb nucleic acid under high salt and low pH value, and separate nucleic acid from nucleic acid under low salt and high pH value, and then move the magnetic beads or transfer the liquid to realize the entire extraction and purification process of nucleic acid. Due to the uniqueness of its principle, it can be designed into a variety of throughputs, which can be extracted from a single tube or 8-96 samples, and its operation is simple and fast. It only takes 10-15 minutes to extract 96 samples, which greatly improves the The experimental efficiency is low and the cost is low, so it can be used in different laboratories, and it is the mainstream instrument on the market.

Principle of Magnetic Bead Method Nucleic Acid Extractor

Magnetic bead method nucleic acid extraction instruments are generally divided into two types: suction method and magnetic rod method:

1. The suction method, also known as the pipetting method, is to extract nucleic acid by immobilizing magnetic beads and transferring liquid. Generally, the transfer is realized by controlling the robotic arm through the operating system. The extraction process is as follows:

1) Lysis: Add lysis solution to the sample, and realize the mixing and full reaction of the reaction solution through mechanical movement and heating, the cells are lysed, and the nucleic acid is released.

2) Adsorption: Add magnetic beads to the sample lysate, mix well, and use the magnetic beads to have a strong affinity for nucleic acids under high salt and low pH to adsorb nucleic acids. Under the action of an external magnetic field, the magnetic beads are separated from the solution. , use the tip to remove the liquid and discard it to the waste tank, and discard the tip.

3) Washing: Remove the external magnetic field, replace with a new tip and add washing buffer, mix well to remove impurities, and remove the liquid under the action of the external magnetic field.

4) Elution: Remove the external magnetic field, replace with a new tip, add elution buffer, mix well, and then separate the bound nucleic acid from the magnetic beads to obtain purified nucleic acid.

2. Magnetic bar method

The magnetic rod method realizes the separation of nucleic acids by fixing the liquid and transferring the magnetic beads. The principle and process are the same as those of the suction method, but the difference is the separation method between the magnetic beads and the liquid. The magnetic bar method is to separate the magnetic beads from the waste liquid by the adsorption of the magnetic rod to the magnetic beads, and put them into the next liquid to realize the extraction of nucleic acid.
