Nucleic Acid Testing of COVID-19

What is nucleic acid testing?

Nucleic acid testing is to find out whether there is nucleic acid of foreign invading viruses in patients' respiratory specimens, blood or feces to determine whether they are infected with the new coronavirus. Therefore, once the nucleic acid is detected as "positive", it can be proved that the virus exists in the patient.

The principle of nucleic acid detection of Novel Coronary pneumonia

All organisms contain nucleic acids except prions. Nucleic acids include deoxyribonucleic acid (DNA) and ribonucleic acid (RNA). The novel coronavirus is a virus that only contains RNA. The specific RNA sequence in the virus is to distinguish the virus from other pathogens marker. After the emergence of the novel coronavirus, Chinese scientists completed the analysis of the entire genome sequence of the novel coronavirus in a very short period of time, and found the specific nucleic acid sequence of the novel coronavirus by comparing it with the genome sequences of other species. During clinical laboratory testing, if the specific nucleic acid sequence of the novel coronavirus can be detected in the patient's sample, it should indicate that the patient may be infected with the novel coronavirus.

Novel Coronary Pneumonia Nucleic Acid Detection Process

The sampling method for nucleic acid detection of the novel coronavirus requires neither surgery nor injection, just a "swipe". At the fever clinic or the corresponding examination room, the doctor will ask the patient to open his mouth and use a swab like a cotton swab to wipe the secretions near the tonsils and pharyngeal crypts (the throat swabs often mentioned in the news) , You can also take the secretions from the back of the nasopharynx through the nasal cavity, put them in a test tube, and then hand them over to the laboratory for the corresponding viral nucleic acid inspection.

Nucleic acid testing requires five steps: sampling, sample retention, preservation, nucleic acid extraction, and on-board testing.

The first step is to collect the secretions of the human body, and wipe the nasal cavity or the posterior pharyngeal wall and bilateral pharyngeal tonsils with a nasal swab or a pharyngeal swab;

The second step requires medical personnel to retain samples, immerse the swab head in the cell preservation solution, and immediately tighten the tube cap after breaking the tail;

The third step is to store the sample tube in a sealed bag and submit it for inspection in time;

The fourth step is to send the required samples to the laboratory for nucleic acid extraction;

The last step is to perform fluorescent PCR nucleic acid detection, and the extract is subjected to fluorescent PCR amplification reaction.
